Does the outlook connector need reinstalling on user PC's after every Zimbra update? - EG: After upgrading from Version 4.5 to 4.53.

Yes it does, the ZCO is also getting bugfixes.

The MSI is "nice" because it replaces the previous version : no need to remove then setup the new ZCO.
