You need to backup the /opt/zimbra directory structure, I'd suggest you start by reading this article and also have a look at some of the other threads on the forums about moving to a new server.

Yes, after the migration to the new server you can upgrade to version 6. This is the easiest way to do it.

thanks for your reply ringnebula, here i got few more question:

1, is there any impact if i upgrade from v5 to v6?
2, is recommended to do upgrade from v5 to v6 after migration or prefer to copy the /opt/zimbra from v5 to v6 dump zimbra server?

1. The upgrade shouldn't cause you any problems. Zimbra has done a good job of making the upgrade process largely seamless.

2. You don't want to do anything with 6 until you have followed steps in the article phoenix linked. That is, copy /opt/zimbra to new server, install same (v5) release on new server, verify it works as expected, upgrade to v6.

I have followed the process above on a few machines and never run into a problem.

You should also make sure that you take a backup before doing the upgrade.
